This month's issue of Nintendo Power has a HUGE amount of info on RE:UC. Here, I'll break it down for you guys.

Who?
There will be a multitude of playable characters in the game. Each location will feature two team members gunning their way through the levels. Here's the list of playable characters:

Chris Redfield (Resident Evil)
Billy Coen (RE:0)
Rebecca Chambers (RE:0)
Jill Valentine (Resident Evil)
Leon Kennedy (RE 2)
Claire Redfield (RE 2)
Carlos Olivera (RE 3)
Albert Wesker (Pictured but not confirmed as playable)

Parts of the story are still being worked on so expect a few secondary characters to be confirmed in the coming months.

How?
Gameplay seems to be similar to your basic light gun game with some pretty revolutionary exceptions. Your aiming reticle has has a orange ring around it acting as an ammo counter. To reload simply shake the remote. All of the environments are fully rendered and destructible. So destructible in fact that if you shoot out too many lights you'll cause a blackout in the area you are in! There will be parts of the game that feature split paths, just shoot the direction you want to go. To regain health you must destroy objects to uncover green herbs. If an enemy gets too close you can kick them like in RE4.

Where?
All of the major locations of the past RE games will be included in UC (With the exception of Code Veronica). Each location will have it's own boss battles and unique enemies and weapons. Major events from the previous games have been re-imagined for UC one of which being the first zombie meeting from the original.


Looks like everything is shaping nicely! Hopefully the game will be fully playable at E3 this year!
